Beschreibung: Collagens are highly conserved throughout evolution and are characterized by an uninterrupted "Glycine-X-Y" triplet repeat that is a necessary part of the triple helical structure. For these reasons it is often extremely difficult to generate antibodies with specificities to collagens. The development of type specific antibodies is dependent on NON-DENATURED three-dimensional epitopes. Collagens are extensively purified for immunization from human and bovine placenta and cartilage by limited pepsin digestion and selective salt precipitation. This preparation results in a native conformation of the protein. Antibodies are isolated from rabbit antiserum and are extensively cross-adsorbed by immunoaffinity purification to produce 'type' specific antibodies. Greatly diminished reactivity and selectivity of these antibodies will result if denaturing and reducing conditions of SDS-PAGE and immunoblotting are used.

Beschreibung: Human Erythropoietin (EPO) is also known as EP, erythropoetin or erthropoyetin, and is a glycoprotein hormone that controls erythropoiesis, or red blood cell production. EPO is a cytokine for erythrocyte (red blood cell) precursors in the bone marrow. is synthesized by renal peritubular cells in adults, with a small amount being produced in the liver. Regulation is believed to rely on a feed-back mechanism measuring blood oxygenation. Constitutively synthesized transcription factors for EPO, known as hypoxia-inducible factors (HIFs), are hydroxylated and proteosomally digested in the presence of oxygen. It binds to the erythropoietin receptor (EpoR) on the red cell surface and activates a JAK2 cascade. Erythropoietin has its primary effect on red blood cells by promoting red blood cell survival through protecting these cells from apoptosis. It also cooperates with various growth factors involved in the development of precursor red cells. EPO has a range of actions including vasoconstriction-dependent hypertension, stimulating angiogenesis, and inducing proliferation of smooth muscle fibers. It has also been shown that erythropoietin can increase iron absorption by suppressing the hormone hepcidin. Erythropoietin has been shown to interact with the Erythropoietin receptor as its mechanism of action within the body. erythropoietin plays an important role in the brain's response to neuronal injury. EPO is also involved in the wound healing process.

Beschreibung: IgA represents about 15% to 20% of immunoglobulins in the blood, although it is primarily secreted across the mucosal tract into the stomach and intestines. LN-5 selectively stains human sebaceous glands in formalin-fixed, paraffin-embedded skin samples. Undifferentiated sebocyte progenitors are negative, and only sebocytes from the onset of their differentiation reveal positive cytoplasmic staining. Since there are very few selective and easy-to-use markers of sebaceous glands, LN-5 antibody can offer a simple and relatively specific way to detect human sebocytes from the onset of their.

Poly-D-lysine hydrobromide, weißes Pulver

Beschreibung: Poly-lysine is a polycation which binds to DNA, red cell membrane and any negatively charged protein. When adsorbed to the culture surface, poly-lysine increases the number of positively charged sites available for cell binding. A compromise between the easier to use lower molecular weight products and the extremely viscous higher molecular weights would be the products in the range of 70,000-150,000.
It is typically used as a coating substrate for culture dishes, slides, etc. It enhances electrostatic interaction between negatively charged ions of the cell membrane and the culture surface. Both the D- and L- form of the poly-lysine can be used as a coating substrate since poly-lysine is a nonspecific attachment factor for cells; however, certain cells can digest poly-lysine. In this case, poly-D-lysine should be used as the attachment factor so that the cells are not disrupted by excessive uptake of L-lysine.
Other uses for poly-lysine have been reported as well:
• Conjugation to methotrexate for increased drug transport.
• Microencapsulation of islets.
• Use in simple reproducible procedure for chromosomal preparations from a variety of tissues.
• Immobilation of living cells in biocompatible semipermeable microcapsules.
• In the preparation of polycationic beads.
• Conjugation to albumin and horseradish peroxidase to enhance cellular uptake.
Poly-D-lysine hydrobromide is a synthetic amino acid that enhances cell adhesion to solid substrates. It has additionally been shown to eliminate prion proteins from infected cells.
