Digoxigenin (DIG) is a steroid found exclusively in the flowers and leaves of the plants Digitalis purpurea and Digitalis lanata. DIG is the steroid component of digoxin, a cardiac glycoside, and can be conjugated onto a variety of biomolecules (proteins, nucleic acids) to be detected in further assays (e.g. ELISA). These antibodies can be labeled with dyes, enzymes or fluorescence, directly or secondarily, for visualization and detection.
DIG has an advantage over biotin due to the fact that biotin is an endogenous molecule of living cells and plays a key role in many reactions, which can sometimes lead to false positive results. Because the blossoms and leaves of Digitalis plants are the only natural sources of digoxigenin, binding of the anti-digoxigenin antibody in other biological material is not a consideration.
