Suspend 30 g of the medium in one liter of distilled or deionised water. Mix well. Heat slowly until the medium is dissolved. Dispense and sterilise at 121 °C (15 lbs of pressure) for 15 minutes.

Tryptic soy broth is used for the cultivation of fastidious and non-fastidious microorganisms. For example, it is used for the isolation and sensitivity testing of all types of pathogens, and for the production of antigens for agglutination and serological tests. Other uses include: urine cultures, blood cultures, cultivation of cerebrospinal fluid, antibiotic sensitivity testing, cultivation of anaerobic microorganisms, vibrios, and Bacteroides, preparation of bacterial antigens, examination of solid foods, tests for bile solubility, qualitative examination of yeasts and molds, with blood the medium becomes richer so that it can cultivate a wider variety of microorganisms.
