Bolton-Hunter reagent (sulpho-SHPP)
Sulpho-SHPP, sulphosuccinimidyl-3 -(4-hydroxypheynyl) propionate, also known as water-soluble Bolton-Hunter reagent, conjugates tyrosine-like functional groups to primary amines to increase the number of tyrosyl groups that can be iodinated by iodine-125 labeling procedures.
- Iodinate before or after coupling to the molecule of interest
- Preserves tyrosine residues that are essential for protein function or immunogenicity
- NHS esters react efficiently with primary amino groups in pH 7 to 9 buffers to form stable amide bonds
Sulpho-SHPP is useful for introducing tyrosyl groups on proteins sensitive to small amounts of organic solvent and for labelling cell surfaces without exposing the cells to membrane permeable solvents.
